Shale Energy Boom Creates Growth, Jobs

Until a few years ago, only geologists got excited about shale. But, today, this type of rock is the source of a new American energy boom, which is creating and supporting millions of jobs in a number of states, including Pennsylvania, Texas, and North Dakota.

Production of oil and natural gas from supported 1.75 million U.S. jobs in just the past few years, according to a report by IHS Global Insight, co-sponsored by the U.S. Chamber?s Institute for 21st Century Energy. Jobs tied to unconventional oil and gas production will reach 3.5 million by 2035, according to the study. Eighty percent of these jobs are on drilling rigs or supply these rigs with pipe, water, sand, and other materials. These shale-related jobs pay more than double the national average. The ripple effect will support the other 20% of jobs?hotel, restaurant, and rail line, for example.

?We?ve known for some time that shale energy is truly a game-changer for America?and now we can prove it,? says Karen Harbert, president and CEO of the Energy Institute.??This study shows that shale energy is already contributing over $200 billion to our economy, with much more to come if policymakers at all levels of government don?t stand in the way.?

Oil extracted from shale has contributed to a more than 25% increase in domestic production of oil over the last four years.?By 2015, oil production is expected to increase by 46% over 2008, and by 2020, there will be a 68% increase?predominantly because of shale.

While U.S. oil production hit a 15-year high in 2012, almost all the increase in production has come from shale development on privately owned lands. The U.S. government continues to restrict development on land that it owns. In fact, in fiscal years 2010?2011, oil production on federal lands declined 11%.

Source of Government Revenue

The IHS study finds that shale energy development is filling the coffers of cash-strapped governments. Government revenues from energy taxes and royalties were expected to reach $62 billion in 2012, rising to $110 billion annually by 2020. A total of $2.5 trillion in additional tax revenue, half going to state and local governments, will be generated by 2035.

?Thanks largely to shale, America now has access to enough natural gas to supply our nation for over 100 years,? Harbert says. ?In addition, shale will reduce the need for oil imports.?In fact, our study shows that by 2020 net oil imports will decrease by 60%, and America will spend $200 billion less on imported oil.?

An Energy Exporter

The shale energy boom has put the United States in the unfamiliar position of becoming a net energy exporter. The Energy Information Administration (EIA) reports that in 2011, for the first time in more than 60 years, the United States became a net exporter of gasoline, diesel, and other refined fuels. EIA says that the United States could become a net exporter of natural gas by 2035.

Energy companies, however, are faced with significant policy obstacles. A 37-year-old federal law prohibits oil drawn from American lands to be sold abroad with some exceptions, including exports to Canada and oil from the Alaskan North Slope.

Now, there is talk in Washington about putting a cap on natural gas exports, even as large increases in domestic supplies have driven natural gas prices to record lows. A study by Michael Levi of the Council on Foreign Relations found that American firms could make up to $3 billion annually by producing and exporting liquefied natural gas (LNG), the form to which natural gas must be converted to be transported overseas.

In May 2012, the Department of Energy approved the first new LNG export permit in years, allowing Cheniere Energy to export natural gas from its terminal in Louisiana. But the agency announced that it needs more time to consider a Sierra Club complaint against Cheniere?s plans. At least 15 other permits are waiting Energy Department approval.

Meanwhile, countries in Southeast Asia, including Singapore and Australia, are building out infrastructure to bring natural gas to market with an eye toward China and Japan. Closer to home, TransCanada Corp. plans to build and operate a $4 billion natural gas pipeline that will send North American gas to eager Asian markets.

Study shows networking properties between brains when guitarists play together

Researchers from the Max Planck Institute for Human Development in Berlin have shown that synchronization emerges between brains when making music together, and even when musicians play different voices. In a study published November 29th in Frontiers in Neuroscience, Johanna Snger and her team used electrodes to record the brain waves of guitarists while they played different voices of the same duet. The results point to brain synchronicity that cannot be explained away by similitudes in external stimulation but can be attributed to a more profound interpersonal coordination.

Scientists working with Ulman Lindenberger at the Max Planck Institute in Berlin already discovered synchronous brain activity between musicians playing the same piece in 2009. The current study goes one step further by examining the brain activity of guitar players performing a piece of music with two different parts. Their aim was to find out whether musicians' brains would synchronize if the two guitarists were not playing exactly the same notes, but instead played different voices of the same song.

To test their hypothesis, the psychologists arranged 32 experienced guitarists in duet pairs, and recorded electrical activity in different brain regions of each musician. They were then asked to play a sequence from the "Sonata in G Major" by Christian Gottlieb Scheidler a total of 60 times, and the duet partners were given slightly different tasks: each musician had to play a different voice, and one of the two was responsible for ensuring that they started at the same time and held the same tempo. Thus, one person took the lead and the other followed.

The duet's brain activities showed coordinated brain oscillations, even when playing different voices of the same duet. Called phase coherence, this synchronous activity suggests a direct neural basis for interpersonal coordination.

"When people coordinate their own actions, small networks between brain regions are formed. But we also observed similar network properties between the brains of the individual players, especially when mutual coordination is very important; for example at the joint onset of a piece of music," says Johanna Snger. The difference between leader and follower was also reflected in the results of the measurement of electrical activity captured by electrodes: "In the player taking the lead, the internal synchronization of an individual's brain waves was stronger and, importantly, was present already before the duet started to play," says Johanna Snger. "This could be a reflection of the leading player's decision to begin playing at a certain moment in time," she added.

The current data indicate that synchronization between individuals occurs in brain regions associated with social cognition and music production. And such interbrain networks are expected to occur not only while performing music. "We think that different people's brain waves also synchronise when people mutually coordinate their actions in other ways, such as during sport, or when they communicate with one another," Snger says.

If You Bring Me a Buyer I Will Pay You 3%-San Antonio TX FSBO Advice from a Realtor?

The well meaning FSBO will let me know that if I have a buyer for their home, they will happily pay me a percentage of the sale..usually 3%. ?I am polite and ask some specific questions about the home, what they have it priced at and how long it's been on the market. ?I will of course ask them why they are trying to sell on their own and what kind of luck they are having. ?

And then, when I get off the phone, that's probably the last time I will even think about the FSBO home. ?Why? ?Well, I am going to tell you a number of reasons why calling an agent and offering them a percentage of a FSBO sale doesn't give you a whole lot of traction nor does it really help to sell your?San Antonio TX home for sale.

First, homes that aren't marketed in the multiple listing service are very rarely seen by an agent that has active buyers. ?Buyers agents send available listings with certain criteria that fits their particular buyer to their clients from the MLS. ?Each buyer has specific things they need looked up,?to include their particular loan type. If your home isn't there, it's not getting sent...

I know that you gave me all the information on your home (that you have knowledge of, do you know if your home will pass a VA or an FHA appraisal?), but that now goes on a piece of paper on my desk and will get buried under a ton of other stuff, probably to be thrown away later. ?Unless I miraculously have a buyer that is looking for your home type at the time you call, I will most likely forget.

Second, homes that aren't marketed period aren't going to be seen by a buyer looking for homes.??Buyers start their search for homes on the Internet and if your home isn't there it won't be seen by them. ? Many times our buyers will alert their agents to homes they have found on a site or two...in fact, this is many times how a buyer finds their agent...while looking for homes that those particular?Realtors??have marketed.

You may have a FSBO sign up in your yard and may have posted a Craigslist add...but trust me, this is not going to be enough to sell your home in San Antonio TX. ?Oh and if you marketed your home on FSBO.com or something like that, buyers looking on those sites are generally looking for a "deal"..are you willing to sell your home as a "deal"? ?

Let's say that you have hired one of those flat fee Brokerages that will put your home in the MLS for you and give you some minor marketing, so you think you have got it covered...

This brings me to my Third Reason...Some San Antonio TX buyers agents do not want to work with a FSBO?to get their home sold even though you have offered to pay their salary. ?As a For Sale By Owner, you most likely (not in all cases) do not know how to negotiate a Real Estate Transaction and although this gives a huge advantage to the buyer and buyers agent, it also may put them at risk and the agent knows that they will be doing all of the work involved to get this to the closing table. ?

And you do know that is an advantage to a buyer and their agent right? ?A buyers agent is going to be representing their buyer and getting the best terms and conditions for them..not you. ?The buyers agent most likely knows way more about the market than you do and has access to the real comparable home sales in the ?past few months..

Zillow and other aggregate sites will not work for you here in Texas as we are a nondisclosure State. ?They don't have the right stats because they don't know what homes have sold for and homes are priced based on actual homes that have sold, not for what homes are currently priced for. ?

So, although you mean well and offering to pay the buyers agent is a nice gesture,?it most likely won't help you to get your home sold. ?My best advice to you is that if selling FSBO is not working for you, (sometimes they do sell, of course you may never know if you got the most you could have), then get with a?San Antonio TX Realtor??to represent you and get your home priced right, marketed expertly and get your home sold.?

House to vote on increasing advanced-degree visas

WASHINGTON (AP) ? A House vote to offer permanent residency to foreign students graduating with advanced degrees in science and math from U.S. colleges and universities is setting the stage for a bigger battle next year on how to redesign the nation's flawed immigration system.

House Republicans, with the help of a minority of Democrats, are expected to prevail Friday in passing the STEM Jobs Act, which would provide up to 55,000 green cards a year to those earning masters and doctoral degrees from U.S. schools in the fields of science, technology, engineering and mathematics.

But the bill is unlikely to go anywhere this year in the Democratic-controlled Senate, and the Obama White House has come out against it, saying it "does not support narrowly tailored proposals that do not meet the president's long-term objectives with respect to comprehensive immigration reform."

A major point of contention is that the bill offsets the increase in visas for the highly educated by eliminating the Diversity Immigrant Visa Program. This year the program made 50,000 visas available to people from countries with traditionally low rates of immigration. About half of those visas go to African nations.

The House voted on a similar STEM Act in September, but it fell short under a procedure requiring a two-thirds majority. It is being revived under rules needing only a simple majority. Republicans are scrambling to show the Hispanic community, which largely deserted them in the recent election, that the GOP is committed to fixing the immigration system.

Earlier this week, two Republican senators introduced their version of the DREAM Act. Their bill would allow young people brought into the country as children without authorization to stay without fear of being deported, an initiative previously opposed by most Republicans.

House Judiciary Committee Chairman Lamar Smith, R-Texas, said the STEM Act, a top priority of the high-tech industry seeking to stop the "reverse brain drain" of highly skilled foreign graduates of U.S. universities leaving for jobs overseas, "will help us create jobs, increase our competitiveness and spur our innovation."

And in an attempt to pick up more votes, Smith added a provision that makes it easier for the spouses and children of residents to come to the United States while they wait for their own green card applications to be approved.

But while most Democrats support increasing STEM visas, there was sharp criticism of the Republican approach.

"This is a partisan bill that picks winners and losers in our immigration system," Rep. Luis Gutierrez, D-Ill., a leader on immigration issues in the Congressional Hispanic Caucus, said of the elimination of the Diversity Visa Program.

"This bill is premised on the dangerous thought that immigration is a zero-sum game," said Rep. Zoe Lofgren. The Democrat, who represents high-tech companies in her northern California district and has long pushed for more STEM visas, said the Smith bill would eventually result in fewer visas issued because far fewer than 50,000 degrees are given every year to foreigners in eligible STEM fields, and the bill does not allow unused visas to be transferred to other programs.

The STEM Act visas would be in addition to about 140,000 employment-based visas for those ranging from lower-skilled workers to college graduates and people in the arts, education and athletics.

The Diversity Visa Lottery Program, created in 1990 partly to increase visas for Ireland, has shifted over the years to focus on former Soviet states and now Africa. In 2010, almost 25,000 visas went to Africa; 9,000 to Asia and 16,000 to Europe. Applicants must have at least a high school education.

Critics say the visa lottery program has outlived its purpose because Africans and East Europeans are already benefiting from family unification and skilled employment visas, and the lottery program is subject to fraud and infiltration by terrorists. Lofgren said it was "preposterous" that terrorists would try to get into a country under a program that picks 55,000 people at random out of more than 14 million applicants.

The provision on reuniting families allows the spouses and children of permanent residents to come to the United States to wait for their own green card applications to be processed one year after applying. The current wait for family members to receive residency is more than two years. The measure says those in the country illegally are not eligible, and family members may not work while waiting for their green cards.

OKLAHOMA CITY ? This is the season of good will to all, a time when people take time to think of others. In addition to giving gifts and planning holiday get-togethers, many people are also dipping into their pockets to make donations to worthy charities. While that?s a good impulse, it?s important to do your charitable giving wisely. The Oklahoma Society of Certified Public Accountants (OSCPA) provides this advice on ways to make your money the most productive.?

Know your goals. When confronted with an urgent call for help from a charitable organization, think first about what other causes are important to you. In fact, before you begin receiving end-of-year appeals, it may be a good idea to decide exactly how much money you have to give to charity, then brainstorm ideas about the causes you care about most. You can?t say yes to every worthy effort, and creating this kind of charity budget will help you focus on the best choices for your donations.?

Be specific. Let?s say you?ve decided you?re interested in charities that help homeless dogs and cats. Before you make your donation, consider whether you have more specific goals in mind. Do you want to give your money to feed or provide medical care for animals in shelters in certain areas? Do you want to support spaying and neutering programs? Do you want your money to be used to support adoption efforts? Determine what your goals are and focus on organizations that have a history of achieving them.

Don?t give to an unknown cause.?Even if a group supports the right cause, there are a number of questions you should ask before you donate your hard-earned dollars to it: How much do I know about this group? Am I confident that it is a legitimate organization that will use my money to benefit the cause it?s espousing? Am I sure that the group will not sell my name and contact information to telemarketers? For more information, look over the organization?s financial statements and consider turning to sources such as Charity Navigator (www.charitynavigator.org), which provides free ratings on a wide variety of charities. Some donors are disappointed to find out how their money is used, with a large amount going to administrative and other expenses rather than to support the cause. Of course, all charitable organizations will have to cover administrative costs to keep operating and pay salaries that attract talented people who can help them achieve their missions. CharityWatch (www.charitywatch.org), which also rates charitable organization, recommends that 60 percent of a donation should go to program services, with the rest spent on fundraising and general administration. Keep in mind, though, that the numbers don?t always tell the full story. If you have a question about how a group spends its money or how well it meets its goals, don?t hesitate to contact them and ask for some perspective.

Understand what?s tax deductible.?Did you spend $200 to attend a charity dinner? If the value of the dinner you received was $75, then you can only deduct $125 for your donation. You must subtract the value of any benefit you receive from a donation before deducting it on your tax return. Remember, too, that no donation is deductible unless you itemize your deductions on your tax return and unless you give to a qualified organization in the eyes of the Internal Revenue Service. (AP) ? Albania celebrated 100 years of independence Wednesday with a military parade, festive cavalcades and an 18-ton cake.

But the celebrations were soured by spats with two of the small Balkan nation's four neighbors, whose officials canceled plans to attend.

Prime Minister Sali Berisha had spoken Tuesday of "Albanian lands" stretching from Preveza in Greece to Presevo in Serbia, and from the Macedonian capital of Skopje to the Montenegrin capital of Podgorica.

The comments were also inscribed on a parchment that will be displayed at a museum in the city of Vlore, where the country's independence from the Ottoman Empire was declared in 1912.

Greek Foreign Minister Dimitris Avramopoulos promptly cancelled his visit to Tirana on Wednesday. A ministry statement said comments like Berisha's "do not contribute to creating a climate of friendship, confidence and good neighborly relations."

Berisha's spokeswoman, Erla Mehilli, said the prime minister was speaking in a historical context and "in no way expressed any territorial claim towards our neighbors in the south, north or east."

Separately, Macedonian President Gjorgje Ivanov called off his visit after Macedonian Prime Minister Nicola Gruevski's car was hit with an egg last week during a trip to Tirana, the Albanian capital.

Serbian official from Kosovo Oliver Ivanovic said: "Greater Albania cannot be created without a war. Greece, Macedonia, Montenegro and Serbia must react to such dangerous aspirations."

Montenegro's president and the prime ministers of Hungary, Croatia and Slovenia turned up, as well as officials from Italy and Kosovo.

Tens of thousands of ethnic Albanians ? some on foot or on horseback ? arrived from Kosovo, Macedonia and Montenegro for the festivities, which started in Vlore.

In Tirana's central Skanderbeg Square, an 18-ton cake was distributed to the public, followed by a military parade.

Berisha's earlier plans to slaughter 2,000 sheep and publicly roast them for the occasion were scrapped after an outcry by animal rights groups.

Hall Of Fame Class Of 2013: Barry Bonds, Roger Clemens, Sammy Sosa Bring Steroid Stigma To Ballot

NEW YORK ? Barry Bonds, Roger Clemens and Sammy Sosa are set to show up on the Hall of Fame ballot for the first time, and fans will soon find out whether drug allegations block the former stars from reaching baseball's shrine.

The 2013 ballot will be announced Wednesday.

Craig Biggio, Mike Piazza and Curt Schilling are certain to be among the other first-time eligibles. Jack Morris, Jeff Bagwell and Tim Raines are the top holdover candidates.

Longtime members of the Baseball Writers' Association of America will vote through next month. The much-awaited results will be announced Jan. 9, with players needing to be listed on 75 percent of the ballots to gain induction.

The upcoming election is certain to fuel the most polarizing Hall debate since career hits leader Pete Rose's betting problems put him on baseball's permanently ineligible list, barring him from the BBWAA ballot.

Bonds, Clemens and Sosa each posted some of the biggest numbers in the game's history, but all were tainted by accusations that they used performance-enhancing drugs.

Bonds is baseball's all-time home runs leader with 762 and won a record seven MVP awards. Clemens ranks ninth in career wins with 354 and took home a record seven Cy Young Awards. Sosa is eighth on the home run chart with 609.

Fans, players and Hall of Fame members have all chimed in about whether stars who supposedly juiced up during the Steroids Era should make it to Cooperstown.

Many of those opposed say drug cheats should never be afforded baseball's highest individual honors. Others on the opposite side claim the use of performance-enhancing drugs was pervasive in the 1980s and 1990s, and shouldn't disqualify candidates.

If recent voting for the Hall is any indication, the odds are solidly stacked against Bonds, Clemens and Sosa.

Mark McGwire is 10th on the career home run list with 583, but has never received even 24 percent in his six tries. Big Mac has admitted using steroids and human growth hormone.

Rafael Palmeiro is among only four players with 500 homers and 3,000 hits, yet has gotten a high of 12.6 percent in his two years on the ballot. Palmeiro drew a 10-day suspension in 2005 after a positive test for performance-enhancing drugs, and said the result was due to a vitamin vial given to him by teammate Miguel Tejada.

Man says prayer group leader told him to kill wife

KANSAS CITY, Mo. (AP) ? Less than three months after he stood as a groomsman in the wedding of two friends he had known since college in Texas, Micah Moore walked into a suburban Kansas City police department and unloaded a dark secret: He had taken the woman's life at the request of her new husband, a charismatic prayer group leader.

Police said Bethany Deaton's death initially appeared to be a suicide. Officers found a note and empty bottle of over-the-counter pain medication along with her body in a minivan parked by a lake on Oct. 30.

It wasn't until Moore confessed nearly two weeks later that police announced she had been killed. He is scheduled for a preliminary hearing on a first-degree murder charge Wednesday.

In the criminal complaint filed in support of the charge, police detailed a stunning series of allegations that Moore made as part of his confession.

Moore, 23, lived with Deaton and her husband, Tyler, in a communal home shared by male members of their prayer group. He told police that several members had sexually assaulted Bethany Deaton and that they were worried she would tell someone. Moore said that's when Tyler Deaton ordered him to kill Bethany Deaton, according to a criminal complaint.

Tyler Deaton has not been charged in his wife's death. Jackson County prosecutor Jean Peters Baker said Deaton was under investigation but declined to elaborate. Deaton does not have a listed phone number and did not respond to requests for comment The Associated Press made through Facebook and phone and email messages to his father.

Moore's attorney, Melanie Morgan, declined to comment.

Tyler and Bethany Deaton moved to Kansas City in 2009 from Texas to attend a six-month internship at the non-accredited International House of Prayer University. The two had met as freshmen at Southwestern University in Georgetown, Texas, in 2005, and two years later Tyler started a prayer group, a former longtime member of the group told The Associated Press on condition of anonymity because he was afraid of retaliation from Tyler Deaton.

Tyler Deaton was listed at one point as a division coordinator for IHOPU's "friendship groups," but the school said that was a mistake. It issued a statement distancing itself from Tyler Deaton after Moore, a student at IHOPU, was arrested.

"Since Bethany's death it has come to light that over five years ago, both she and Mr. Moore joined an independent, close-knit, religious group in Georgetown, Texas," the school said in a statement. "This religious group of fewer than 20 people was led by Tyler Deaton. They relocated to Kansas City over the last few years and operated under a veil of secrecy."

IHOPU is the educational arm of International House of Prayer of Kansas City, an evangelical Christian group focused on missions and preparation for the end of time.

The Deatons' prayer group had at least two houses, with women living in one and men in another. Bethany Deaton, 27, moved into the men's house with Tyler Deaton after they married in August.

According to the criminal complaint, Moore told police that men in the house began drugging Bethany Deaton and sexually assaulting her soon after she moved in. He said she was seeing a therapist and group members became concerned she would tell the therapist about the assaults.

Moore and other men who lived in the house told police that several group members also were having sexual relations with Tyler Deaton, unbeknownst to his wife. One man, whose name was blacked out of the criminal complaint, told police that Tyler Deaton said after Bethany Deaton died that he had had a dream he killed his wife by suffocating her.

Moore told detectives Tyler Deaton instructed him to kill Bethany Deaton because he knew Moore had it in him to do it, and that Moore reported back to Tyler Deaton after she was dead. Moore told police that he had placed a bag over Bethany Deaton's head and held it there until her body shook.

Health-care providers can play critical role in reducing and preventing intimate partner violence

ScienceDaily (Nov. 28, 2012) ? In a perspective article to appear in the Nov. 29 issue of the New England Journal of Medicine, researchers from Boston University Schools of Medicine and Public Health (BUSM and BUSPH) report that health-care providers can play a critical role in helping to reduce and prevent intimate partner violence (IPV) by screening and referring patients to appropriate resources.

The U.S. Centers for Disease Control and Prevention recently released a comprehensive report on the prevalence of sexual violence, stalking and IPV in the U.S. The report relays the alarming findings that 35.6 percent of women in the U.S. are raped, assaulted or stalked by intimate partners at some point during their lives, and approximately six percent experience these events in a given year. Men are also at risk: 28.5 percent report lifetime victimization and five percent report past year victimization. The annual health care costs for women who are experiencing ongoing IPV are 42 percent higher than those for non-abused women, and it has been estimated that the cost of providing health care to adult survivors of IPV ranges from $2.3 billion to $7 billion in the first year after the assault.

Now that the Affordable Care Act requires insurance coverage to include domestic violence screening of women, physicians are taking a new look at how they respond to patients who are domestic violence survivors. There are several steps doctors should take when patients report potential IPV, including acknowledging the admission of abuse. "We advise thanking the patient for trusting the provider with the information, and expressing concern about the patient's safety," explained lead author Jane Liebschutz, MD, MPH, FACP, an associate professor of medicine and social and behavioral sciences at BUSM and BUSPH respectively.

Other steps include: asking the patient if he or she would like to be connected to IPV advocacy services; offering the patient the National Domestic Violence hotline number; considering whether child protective services are required and screening the patient for co-morbid depression, anxiety and substance abuse.

According to Liebschutz, when a patient screens negative for IPV, but providers nevertheless suspect that they are experiencing abuse, it is important that the provider not force disclosure. "It is not critical that the patient acknowledges IPV victimization in order to receive a benefit from the screening. Having asked IPV-related questions signals to the patient that the provider is caring and concerned, trustworthy and willing to discuss the topic during a future visit," she added.

Finally, providers need not receive a positive screening response in order to provide universal education about IPV. "Even if a patient screens negative, providers are encouraged to state that many patients do experience IPV at some point, and that there are many resources to help people who feel unsafe in their relationships," said co-author Emily Rothman, ScD., an associate professor of community health services at BUSPH.

Finding may lead to treatment that protects against autoimmunity

BOSTON November 28, 2012 As part of their ongoing research on the role of genes in the development of type 1 diabetes, Joslin Diabetes Center scientists, in collaboration with scientists at the University of Wrzburg, have demonstrated how a genetic variant associated with type 1 diabetes and other autoimmune diseases influences susceptibility to autoimmunity. The findings appear in the upcoming issue of Diabetes.

Recent studies of the human genome have identified genetic regions associated with autoimmune diseases such as type 1 diabetes. Joslin scientists in the Section of Immunobiology seek to understand how genes that are most widely associated with various autoimmune diseases contribute to disease risk.

One of these genes is PTPN22, which plays a role in lymphocyte (immune cell) function. A PTPN22 variant (or mutation) has been implicated as a risk factor for type 1 diabetes and several other autoimmune disorders. PTPN22 is involved in the formation of a key protein known as lymphoid tyrosine phosphatase (LYP), which helps control the activity of T and B cells in the immune system. The PTPN22 mutation generates a variation of LYP with a different molecular structure.

Most studies of the PTPN22 disease variant have suggested that this variant is a gain-of-function genetic mutation that enhances LYP activity and lessens the activity of T and B cells, which increases susceptibility to autoimmunity. "When immune cells are less reactive during the maturation phase of their development, the cells can evade mechanisms that help protect against autoimmunity," says study lead author Stephan Kissler, PhD, of the Section of Immunobiology. However, one study which analyzed data from humans and genetically modified mice suggested that the LYP variant associated with type 1 diabetes is a loss-of-function mutation that reduces LYP activity.

To help resolve the conflicting data, Joslin scientists conducted studies with a unique mouse model developed by Dr. Kissler's graduate student and co-author, Peilin Zheng. Using a technology that combines RNA interference, a method to silence gene expresson, with lentiviral transgenesis, a method to genetically modify animals, the scientists can manipulate gene activity in the most widely used mouse model for type 1 diabetes, the nonobese diabetic mouse (NOD). In this study, the researchers were able to easily turn off and on the PTPN22 gene in the NOD mouse. "We are the first to use this approach in the NOD mouse model," says Dr. Kissler. "It provides a very powerful way to study the contribution of PTPN22 to disease."

When PTPN22 was turned off in mice, mimicking a loss-of-function mutation, the researchers observed an increase in regulatory T cells and a decreased risk of autoimmune diabetes. "This is the first study conducted on the diabetic mouse model that supports the LYP gain-of-function hypothesis," says Dr. Kissler. "Our work should help to resolve the controversy."

By providing additional data that suggests the potential therapeutic value of PTPN22 manipulation, the study may further the development of new therapeutic options that inhibit LYP to reduce or prevent autoimmunity. "Our goal is to treat autoimmunity. Inhibiting LYP in patients may increase regulatory immune cells and could confer protection against autoimmunity, but it remains to be tested if our promising findings in this mouse model are reflected in humans," says Dr. Kissler.

The Joslin scientists are following up on this study to deepen understanding of how inhibiting PTPN22 affects T and B cells.

Report: Microsoft Prepping 'Xbox TV' Set-Top Box

Over the years, the Xbox 360 has morphed from a gaming console to a full entertainment experience thanks to the addition of streaming video and other apps.

According to The Verge, Microsoft plans to spin off the entertainment component of the console and introduce a set-top box that is basically an Xbox 360 without the more serious gaming option.

Instead, it would focus on movie and TV show streaming, as well as casual gaming and other apps supported by the platform.

This "Xbox TV" device will be introduced next year, the blog said, and might include deals with TV makers to add the Xbox software to Web-connected TVs. Presumably, consumers could pick up an Xbox TV for a price that's cheaper than the full Xbox 360 console.

In line with Microsoft's current move for a total Windows experience, The Verge said the effort might allow for the full Xbox experience on a Windows Phone.

Microsoft would not confirm the report, telling The Verge only that it is "always thinking about what is next."

The news comes amidst reports that Apple is working on a similar service. In a note to investors, Jefferies analyst James Kisner cited a source within an unnamed, North American cable provider who told him that the cable firm was "working to estimate how much additional capacity may be needed for a new Apple device on their broadband data network." A Piper Jaffray analyst, meanwhile, thinks Apple will introduce an actual TV set next year.

From Canada, a tough economist for Bank of England

TORONTO (AP) ? Naming the first foreigner to serve as governor of the Bank of England since it was founded in 1694 might have been expected to cause a fuss. Yet the appointment of Canadian Mark Carney has won bipartisan praise in Britain.

That's because few people in the world have Carney's qualifications.

The Bank of Canada governor is a highly-educated economist with Wall Street experience who is widely credited with helping Canada dodge the worst of the global economic crisis. He gained a reputation along the way as a tough regulator who stood up to JPMorgan Chase CEO Jamie Dimon.

Moving to the top job at Canada's central bank just before the global financial crisis hit in 2008, Carney slashed interest rates to historic lows and was the first central banker to commit to keeping them at a historic-low level for a definite time, a step the U.S. Federal Reserve would follow.

Canada was spared a mortgage meltdown or subprime lending crisis, and its banks, more regulated and less leveraged than Europe's, are rated among the world's soundest. Britain, however, has endured a decade of economic turbulence, and seemed ready for a foreigner to shake up its central bank.

Besides his job as a central banker, Carney, 47, also heads the G-20's Financial Stability Board, a global supervisor of regulatory changes in the financial system. Carney's championing of stiffer regulation was highlighted in a run-in last year with Dimon. The Bank of Canada chief stood his ground in a closed-door meeting in Washington when Dimon directed a diatribe toward Carney over what the JPMorgan Chase chief called "anti-American" global reforms against banks.

"When there was pushback on the regulatory front he dug in his heels and made his case that, no, we shouldn't slow down, we should be pushing forward," said Craig Alexander, a Toronto-Dominion Bank economist.

"This is the sort of thing that makes him qualified to take on the challenge of the Bank of England because he's going to be materially involved in shaping regulatory reform of the financial system in London," he added. "So the fact that he would stand up for the regulatory regime is exactly the characteristic one would be looking for."

The transnational appointment is a mark of today's globalized world, while also echoing a past in which Canada was a part of the British Empire.

Like other central bankers, Carney is a former Goldman Sachs executive. He worked for 13 years in London, Tokyo, New York and Toronto, before being appointed deputy governor of the Bank of Canada in 2003. He has both financial industry and government regulatory credentials.

"He looks and sounds the part," CIBC World Markets economist Avery Shenfeld said. "He has the academic background, the gravitas and the experience to withstand the fray of central banking in a city like London."

Carney, from the remote northern town of Fort Smith in the Northwest Territories, gained a bachelor's degree in economics from Harvard University in 1988, and master's and doctoral degrees in economics from Oxford University. Like many Canadians, he played ice hockey, serving as a backup goalie for Harvard.

He lived in London for a decade and his wife and children are dual nationals. British Treasury chief George Osborne, announcing the surprise choice to the House of Commons on Monday, said Carney will apply for British citizenship. He takes up the new position July 1, succeeding Mervyn King, who has headed the bank since 2003.

Before Monday's announcement the frontrunner for the job was widely seen to be Paul Tucker, the bank's deputy governor. But Tucker was likely bypassed because of his embroilment in the Libor-fixing scandal. In July, he was forced to testify to lawmakers after emails emerged showing he had communicated with Bob Diamond, the ex-chief of Barclays, over the setting of the inter-bank interest rate.

Carney's appointment won bipartisan praise in Britain. Ed Balls, the opposition Labour Party's economics spokesman, called it "a good choice," while Osborne said Carney was "quite simply the best, most experienced and most qualified person in the world to do the job."

Although Osborne said it was the first time the Treasury advertised the governor's job, Carney said he hadn't applied. "I didn't apply as part of the formal process to the position. These discussions really only intensified in the last two weeks," Carney said at a news conference in Ottawa.

Carney said he initially turned down the job but British officials kept after him. He said he changed his mind because his term as governor of the Bank of Canada was ending soon, the London job was a big opportunity and the fate of the British economy was critical.

"It's very important for the global economy that the U.K. does well, that it succeeds in this rebalancing of their economy, that the reform of the British financial system is completed, " he said.

Carney said he would take the job for five years instead of the usual eight, and many Canadians are speculating that Carney may return to Canada to run for prime minister. The opposition Liberal Party of Canada had eagerly courted him, but Carney said this month he would remain a central banker for some time.
